depends on want say. commas can alter intent of sentence.
1) missing you, today. - means miss person more typical amount on particular day.
2) missing especially, today. - "especially" modifies missing, instead of 'today', therefore placing focus on how special 'missing' is, while downplaying sort of significance day itself.
i'd use comma if want obliterate ambiguity between 2 options.
@ mr micellano (or spelled): syntactically speaking, it's not sentence fragment. there subject , predicate, subject imbedded informally in word "missing" which, based on operation of syntax, contains "i am" within it. advice on using hyphens, or fretting on other bizarre modes of punctuation, therefore incorrect.
